Which is particularly beneficial for individuals with xerostomia. It basically acts like a key that fits into specific locks called muscarinic receptors on the surface of salivary gland cells. Once pilocarpine binds to these receptors, it triggers a series of events that lead to increased saliva production. A simplified breakdown of the mechanism of action could be: 1. Pilocarpine attach to muscarinic (M3) receptors located on the outer surface of salivary gland cells. 2. G protein activation: Upon binding, pilocarpine triggers a conformational change in the muscarinic receptor, activating a G protein (specifically Gq) coupled to the receptor. 3. Phospholipase C activation: The activated Gq protein stimulates phospholipase C, an enzyme that breaks down phosphatidylinositol 4,5-bisphosphate (PIP2) into diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). 4. Calcium mobilization: IP3 diffuses into the endoplasmic reticulum, where it triggers the release of calcium ions. 5. Protein kinase C activation: DAG activates protein kinase C (PKC), which is involved in various cellular signaling pathways. 6. And finally…. Saliva production and secretion: The increased intracellular calcium concentration and PKC activation trigger a cascade of events that lead to saliva production and secretion. These events include the activation of aquaporins (water channels) and exocytosis of secretory vesicles containing salivary components But the as it affects salivary glands, it can also affect other exocrine glands, including sweat glands and lacrimal glands.
